Enzymatic Preparation and Antioxidant Activities of Protein Hydrolysates from Allomyrina dichotoma Larvae

Abstract
Allomyrina dichotoma larvae (ADL) extracts have been reported to possess multiple biological activities; therefore, this study was conducted to evaluate the antioxidant properties of ADL protein hydrolysates. Protein hydrolysates were prepared from ADL powder by enzymatic hydrolysis using five different proteases (alcalase, bromelain, flavourzyme, neutrase, and papain). Evaluation of the peptide contents and SDS-PAGE analyses revealed that ADL treated with alcalase, neutrase, and flavourzyme showed a high degree of hydrolysis (HD), whereas the HD value of those treated with bromelain and papain was minimal. The RC50 values of the protein hydrolysates (<3 kDa) obtained from three different antioxidant analyses revealed that they had similar antioxidant activities. In addition, the alcalase hydrolysate showing the highest production yield of low molecular weight protein hydrolysate was further tested for its inhibitory effect on peroxidation of linoleic acid by measuring the thiobarbituric acid values and the results showed that treatment with the alcalase hydrolysate (100¡­400 ¥ìg/mL) significantly inhibited linoleic acid peroxidation during five days of incubation. These findings suggest that protein hydrolysates from ADL represent potential sources of natural antioxidants.
